In an approach to optimize substrate specificity, two amino acids have been substituted at the cleavage site of the collagenase substrate, Dnp-Pro-Leu-Gly-Leu-Trp-Ala-D-Arg-NH? (M-1855)· The substrate possesses a 10-fold better kcat/Km value for human fibroblast collagenase than the FRET substrate M-1855· However, monitoring of the enzyme activity using M-1905 requires HPLC analysis as the quencher (Trp) has been removed by the amino acid substitutions
